  • how to check the primers specificity in a local computer

    Could someone please tell me how to check the specificity of primers in a local computer? I know there are lots of online tools available, i.e. primer-blast. However, the database of my research model is not updated in ncbi or any other database. Now I have a better database available in my local computer and was trying to set up a local primer-blast system. But I cannot find the download link of primer-blast or any replacement softwares to check the specificity of my primers with my local database. Could someone please shed light on this?

    If you are looking to confirm specificity of the primers you have already designed then you can use blat locally (https://genome.ucsc.edu/FAQ/FAQblat.html). You will need a unix machine. With blat a multi-fasta file containing your genome sequence can serve as the "database".
