  • Covaris 96 well format

    I am carrying out a large scale protect soon where I will need to prepare many libraries. We are planning to use a Covaris for shearing, but only have access to a instrument that does one sample at a time. We also need to pay for the use which is quiet expensive.

    Does anyone know if there is a service available anywhere have samples run on an E series covaris with 96 well capability?

      Thanks for your reply, I have found out that here in the UK the Covaris supplier, Kbioscience now offers a shearing service.

        depending on cost, you might also consider NEB fragmentase. $2/sample and easy to scale up

          Thanks, we are trying to cost up the different methods and assess the pros and cons of each. Have you worked with the enzyme methods? Is there a problem with non-random cleavage?

            i prepared samples with enzyme fragmenting, we have not sequenced so i can't comment about non-random proclivity but it was validated for wgs. the yield is lower than covaris.

            we are working on large sample number exome resequencing and we are very happy with the other enzymes from NEB for illumina libraries. using our own adapters and oligos reduced our sample prep to near $50/sample.
