  • Data Storage Space NGS

Can anyone please help me with my inquiry.

I will have a next-generation sequencing data coming from external source. How much will be data size on disk (in Gb) for a next-generation sequencing dataset. An estimate will be fine as we want to decide on storage space to have for this.

    Mouse Samples - 400
    Platform - illumina
    Single end

Anything else you want to ask please let me know. Also for analysis using bowtie/cufflink and other standard tool, how much additional space is advisable.

It very much depends on your downstream analysis.
Raw data currently runs between 0.5-1 GB of (compressed) sequence for a single illumina lane, but is still increasing.

You need at the very least that much again for the aligned data etc, and more for further processing. Not to mention temporary disk space. My 'worst' project runs around a factor of 8.5 on top of the raw data.

Just keep in mind, storage is not just raw disk space - you want redundancy, backup and checksums on your data, and it will always be cheaper to buy the same amount of storage next year.

      Strategies for Sequencing Challenging Samples
      by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
