  • MiSeq/BaseSpace Stream

    Hi,

Anyone know what a BaseSpace data stream going from a MiSeq to BaseSpace looks like? Would it possible look like a torrent? Our Institution recently made some change to the ports filtering and I'm suspecting that it has inadvertently blocked our upload.

    Thanks,
    Andor

Here is a quote from Illumina about basespace and required ports.

Contact your local IT administrator if local security policies have to be modified to allow access to BaseSpace. BaseSpace uses SSL/https port 443 and the domains *.basespace.illumina.com and *.s3.amazonaws.com. Data streaming to BaseSpace is encrypted using the AES256 standard and uses SSL for protection. More information on encryption can be found at http://blog.basespace.illumina.com/2...pace-security/

Apparently, there's no blocking...but the stream is being blocked. Betcha the University thinks it's an undergrad up/down-loading movies.

We have had trouble with firewall and amazon AWS (many Illumina websites are hosted on AWS). Amazon seems to change IP's on AWS sites (multiple times a day, possible security measure). Unfortunately amazon AWS uses many subnets so if your firewall uses IP's to manage access then all those subnets would have to be allowed through.

Ask your firewall admins as to how they are handling outgoing connections on 443.

            Current Approaches to Protein Sequencing
            by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

            Strategies for Sequencing Challenging Samples
            by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
