Here's the script, Hope it works.
Just to clarify - I was given the 454 data in fasta format, not SFF.

I'd be interested in hearing how things go for you.

the output doesnt seem to be correct , can u check it up with the min bases above?

the problem is with the lower case bases. change all bases to capital letters and then try

How many reads are you giving velvet?
The program itself is limited in the number of reads it can handle, even if your machine isn't.
I'm working on the cluster located on our solid 3 machine. not optimal, but an ok temporary solution.

                  Dear Temima,

I wonder whether you have found a solution with the denovoadp problem. I am running into the same problem "reads match to negative position".

                  Best,
                  Ioannis

                    afraid not

SOLiD support suggested a new tool called Cortex which is supposed to be uploaded to sourceforge soon. So far, it's not there.

Framework for individual and population genome assembly and discovery of genetic variants from sequence reads.

Essential Discoveries and Tools in Epitranscriptomics
                        by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist on Modified Bases...

                        Current Approaches to Protein Sequencing
                        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...
