  • barcode library for ion torrent

Hi everybody, I am new in the NGS field. I want to ask a question. If I want to barcode my samples, should I do paired barcode or one side barcode, considering the reading length of ion torrent, If the fragment is more than 300bp, maybe there will be an issue to do paired barcod?

I want to do a pilot experiment to test my propose. but I hardly find literature regarding to my question.

I wonder if any of you have experience on this, can you share your thoughts?

    Greatly appreciated!

One barcode because Ion Torrent software doesn't look for barcodes on the other ends.

      thanks for your reply,
Another question is, I am not sure in which end to put barcode, in the beads end, is it P1 adapter and the other is A adpater? If I want to add barcode for sucessful read through, which end is better.

      Thanks a lot

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
