I recently performed a ChIP with the intention of fragment library construction and sequencing on the 5500. I am using the INvitrogen MAGnify kit.
When I performed my qPCR and analyzed it using the percent input method, I only achieved a signal of 2% for a ChIP using 5 ug H4 pan acetyl polyclonal from Active Motif. I use chromatin equivalent to 100,000 cells. I also run an IP with a well-validated H3K9 polyclonal as a positive control for the IP. My source of chromatin is neutrophils isolated from whole blood of a healthy donor.
Anyone have any experience with the % input signal from a ChIP using and H4 antibody?
