  • High-Throughput DNA Shearing

    Hello,

Does anybody have any experience or recommendation about DNA shearing tools that can handle multiple samples simultaneously? I know there are some systems out there that can sonicate a whole plate. Has anybody tried them? Also, I have seen several posts on the Bioruptor: does anybody know if it can go down to 150-200bp peak size? How long does it take?

    Thank you so much!

We are working on a way to fragment and add adaptors using a transposon-based method. Our fragments tend to smear in the 150-2000 bp range. How critical is it for you to be in the 150-200 bp range? We can gel purify, but would like to eliminate this step if possible. Which adaptors do you need to add?

        Piranha

        Hi,

I believe ABI is working on a 96 well sonicator for SOLiD called a 'Piranha'. I heard something about it when I attended a SOLiD V3 launch seminar. Talk to your rep and see if this is the case.

          Current Approaches to Protein Sequencing
          by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

          Strategies for Sequencing Challenging Samples
          by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
