  • Error in maker (gene prediction)

    Hello Everyone,

Does anyone know about below error in maker annotation :
    ERROR: Non-unique top level ID
While this is technically legal in GFF3, it usually
indicates a poorly fomatted GFF3 file (perhaps you
tried to merge two GFF3 files without accounting for
unique IDs). MAKER will not handle these correctly.

I used fasta file only with one contig. I don't know why I am getting Non-unique ID error.

If anyone know please reply. I am waiting for responses.
    Thanking you in advance!

    Regards,
    Rajesh

It seems you have included a GFF3 file in your maker options - one of your conf files. I would recommend supplying evidence as FASTA files to annotate your genome with, so make sure all the GFF3 files in the options are commented out.

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
