  • Strange migration in fragment analysis

    Hi
Since few days we have strange migration on our ABI 3130.
We change the capilary and it was the ok for two days then the problem came back.
I have reads lots of troubleshooting guides but nothings comparable.....
Every consumable were changed but the problem still there.
Is anyone of you have a idea of this problem type?
In attachement an exemple of LIZ500 migration (capillary 2 and 3 presents strange migrations)
Thanks a lots for your help

    Hello,
try changing the polymer, also refill the array with the new polymer. Check for bubbles in the fluid system. What about the room temperature? We see those problems more often, if the room temperature is to high or vary to much.

    Hope that helps a bit.

      Hello Fin,

We have changed also the reagents. New lots of polymer, buffers, HiDi, LIZ500, but no amelioration.
The température is 24°c in the lab and is stable +/-0.5°c during the last week.
      Thanks for your help.

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
