Going to 300, or going to 2x150? The two aren't the same thing.
But yes, 2x150bp with an overlap gives excellent results. In fact with an insert size of 450bp I've seen the Illumina give better assemblies than 454 can manage.
The cost differential is so large, even if 454 can give better results, many users of our service aren't opting to pay the difference. -
Ah, as soon as we get and figure out how to squeeze MiSeq I'd like to try PE 250 on 400bp fragments. 5-10M reads.
