  • Maximum Insert Size of Illumina Libraries?

    Hi, all:
    I'm wondering what is the maximum insert size of illumina paired-end or mated-paired libraries? Some source saids it's 20kb or 40kb, I'm wondering if it is possible that I can get a even longer insert size?
    Moreover, will library preparing process become more complicated or harder when the insert size increases?

    If you are talking about mate pair, there are people that can go up to 40kb+. This is not something the average user will be able to accomplish.

      I was wondering if you can tell me what is the potential technical difficulties when constructing libraries with ultra-large insert size?

        Does anyone have final libraries which show shoulder peaks on their larger insert libraries?
