    From the RFP:

    The U.S.Army Medical Resarch Institute of Infectious Diseases would like to acquire a nucleic acid template detection assay called Heterologus Elongation Ligation Reaction and Sequencing (HELSR) that has several advantages over library preparation technologies currently available. The HELSR probes differ from PCR primers in that they recognize targets on the same strand and barcodes that are used that are specifically resilient to failure with Illumina and IonTorrent sequencing errors. This technology allows the recognition of target template and the formation of the barcoded ligation product only in the presence of template and this critical step is separated from an amplification step. The result is a library that contains only the intended targets to be amplified greatly increasing signal to noise. Specifically analysis software is used to demultiplex large sequencing runs consisting of hundreds of samples and tens of millions of reads and output the results in as little as two hours. Hundreds of probe sets can be screened at a reduced cost allowing us to test many more possible target probes and thereby maximizing the efficiency of probe performance in the final assay.
