Hi, everyone. This is my first post here. I'm new to GAII sequencing. Here is my question:
Can I mix small RNA sequencing primer and genomic DNA sequencing primer in one lane while synthesis of the first-base? Because this lane contains two samples require different sequencing primers. Thanks. |

how would you separate the two samples?

by adding a "barcode" at the 5' end of the library, so that I can separate the two samples after sequencing.

As long as you can tell the samples apart, there is no issue. Illumina v2 CBOTs kits have a cocktail of primers, including the genomic and small RNA primers in the same lane.
