storage of sheared DNA samples overnight

Hi

I am a new user with Covaris. I build DNA sequence libraries from clinical samples. I extract DNA from the samples and shear them to sizes 150 - 200bp and use these fragments to build sequencing libraries (using illumina multiplex sequencing kit). After shearing on covaris, is it advisable to store the fragments overnight and build library the next day? If it is alright, are there any precautions or things to take note of in storing them?

I don't foresee any issues with doing this other than the obvious ones. For example, if your DNA is not purified of contaminating DNAses prior to sonication, then it may degrade while being left overnight. But, if you are not seeing any issues with degradation prior to sonication, you are probably fine.

I have done this as well. It's probably not the best practice for sheared DNA, but I have not personally seen catastrophic degradation.

Overnight at 4 degrees should not be a problem.
