  • mid-range inserts for HiSeq de novo genome sequencing

    Hello. I wonder if any of you is familiar with the protocol to generate paired-end DNA library for illumina HiSeq in de novo genome sequencing.

    I did paired-end sequencing with 200bp inserts, and want to try with longer inserts (e.g., 800bp, 2kb) to get higher continuity of assembly.

    At PAG meeting in San Diego early this year, I heard about a series of libraries with different insert lengths used in the Parrot genome project, but have not found any publication and technical information for that project.

    I would also check out this post, where long inserts have been shown to still sequencing. This indicates that 800bp inserts should be fine (certainly on MiSeq).
