I'm in a position where I need to advice biologists which technology is suitable for their experiment purpose. Is there any study which explores the ideal condition of reads (or rule of thumb) for different NGS applications? for example, for de novo sequencing we need X coverage of single-end / paired-end reads with at least Y bps reads to achieve the best result.
