  • rRNA-depleted RNA profile looks weird

    We received these bioanalyzer profiles after rRNA depletion (using Illumina stranded Total RNA kit, we check for removal of rRNA even though it's not in the protocol) from Th17 cell total RNA. The "normal" picture attached is of samples from same experiment but earlier time points. The last two times points though are showing left-shifted profiles for the control and right-shifted profiles for the KO. All total RNA profiles looked good with >8.4 RIN values. Have any one seen this type of profiles before ("abnormal" picture).
    The Total RNA was DNase I treated, so genomic DNA cannot account for the right-shifted profiles. These (normal and abnormal) were all processed with the same reagents from a Illumina Total RNA kit on the same day. Any thoughts are much appreciated!!
