Hi All,
Has anyone had any experience trying to add UMI to the smart-seq2 protocol? I've done some searching, but I haven't found an account of anyone doing so. I would like to know if it is even possible. Thank you! |

For this protocol UMI can be added to RT primer and SMART oligo. But they will be lost once amplified cDNA is fragmented/tagmented. With some modifications it is possible to keep primer and SMART oligo UMIs but reads from main body of transcript (except termini) will not have UMI.
