We are planing a miRNA sequencing for our mouse muscle samples.
Usually, we get very good RNA samples with our Qiagen miRNeasy kit, with RIN >8.
Unfortunatelly, for this batch, we have RIN ~5.0 for all samples.
A260/A280 is always around 2.0, which excludes contamination. The tissue is freshly dissected and stored in RNAlater at -20 C for 2 days. Then we extracted RNA using Qiagen miRNeasy kit.
It is some weird that we have such low RIN numbers.
Now, we need make a choice, to throw these RNA samples away or go forward for microRNA sequencing?
Anyone has similar experience?
Advice is appreciated.
