  • Genomic enrichment on non-model organisms...

    Hi all,

Just wondering if anyone could give some advice about the following. We would like to enrich for fosmid end sequences in a fish (nonmodel) and shrimp species, for subsequent high coverage sequencing (454 or SOLiD) in a number of individuals. My understanding is that Nimblegen arrays are restricted to human and mouse at the moment, but Agilent technology is more flexible. Can anyone suggest what would be the most suitable technology for this type of enrichment...if it is indeed feasible?

    Thanks,
    Matt

    Hi Matt
You can order custom sequence capture arrays from Nimblegen for any species and do the enrichment yourself, the service they provide is only for human/mouse. I have just tested it and it works great with Solexa. The arrays cost about 600$ and you order minimum of 5. The expensive thing is that you need to have the MAUI hybridization station to perform the hybridization.

      Hi sigusn,

Thanks for the quick reply! Good to hear Nimblegen can provide custom capture arrays, I guess I glossed over their website a bit too fast...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
