  • Experience with Illumina TruSeq kit for NGS

    Hello everyone,

    I am considering purchasing Illumina's TruSeq kit for multiplex sequencing. Does anyone have any experience with this? I am wandering if its very complicated and if it takes a lot of time to get experienced with it. Is it difficult to get good results with it? How many samples does it approx. take to get 'the hang of it'?

    Can anyone give advice on this?

    I prefer the NuGEN Encore multiplex kit. The protocol is easy and gives good libraries. It only takes 3 hours to do too.

      The TruSeq kit works well (i.e., produces diverse libraries, gives high quality sequence reads, demultiplexes without significant sample loss), but takes longer than the NuGEN kit. There are a number of manipulations but, if you have experience with molecular biology, the protocol is not difficult.

        The TruSeq kit is also a LOT cheaper... at least, it is with our Illumina pricing in Australia (YMMV).

          We've made more than 1200 library since early this year using TruSeq. Very robust and good quality for Illumina sequencing. Much simplified compared to prior version of library kit.

            I have just finished some trials with the Truseq protocol, it is quite easy to use (once you have some experience with molecular work as HESmith says), could be done in a (long) day, ~10 hours; first few runs take 2 days to go through it more slowly.
