Default Minimum number of samples for RNA-Seq

A reviewer recently asked to define a RNA-Seq study as pilot since we had 32 samples (16 patients each in two comparing groups). Please advise what should be a statistically sound minimum number of samples to be considered for transcriptome studies when comparing two groups?

The absolute minimum is 3 per group. The "pilot" request is presumably due to the fact that an n of 64 is well within the range of a pilot study for clinical trials. Presuming you're not doing a clinical trial, it'd be appropriate to simply remind the reviewer of that.
