  • Improving PCR-free NGS library preparation

PCR-free NGS libraries reduce bias, improve coverage of difficult areas, and enhance the detection of sequence variants. However, the current PCR-free NGS library prep kits tend to have low sequencing yields than that of PCR amplified libraries because:

    • Full libraries are mixed with partially ligated libraries and un-ligated DNA fragments, thus making it difficult to optimize library quantity due to inaccuracies in library size and amount.
    • Partially ligated libraries and un-ligated DNA fragments reduce cluster efficiency.

In order to address this issue, BioDynami has developed the PCR-free NGS DNA Library Prep Kit.

Our PCR-free kit effectively removes partially ligated libraries and un-ligated DNA fragments, resulting in only fully ligated DNA fragments in the final library. The maximal sequencing yield can be reached since cluster density is optimized based on accurate library sizes and amounts. Also, there is no clustering interference caused by partially ligated libraries and un-ligated DNA fragments.

Features of BioDynami PCR-free NGS kit:

    • Pure, full library: Contains only fully ligated DNA fragments
    • Fast: Hands-on time around 10 min; Total time less than 2 hrs
    • Simple workflow: Two reaction steps for library construction; Simple reaction setup with master mix solution for reaction steps
