  • RIN - Drosophila RNA Pico Bioanalyser

    Hi All,

    Anyone please let me know how to calculate RIN number for Drosophila RNA Pico Bioanalyzer data.

    I did not find a reference RNA pico Bioanalyzer profile for Drosophila tissue.

    You can't. RIN is calculated from the ratio of 28S to 18S rRNA. However, fly 28S is processed into two fragments of nearly equal size, which are also the same size as 18S.

      The RIN value is basically just a voodoo number created by Agilent to quantify RNA integrity. Checking the electropherogram and gel-like image from the Bioanalyzer is all you need to determine whether your RNA is intact. If you are in doubt post the traces here- although we don't all know exactly what intact Drosophila total RNA should look like many of us are familiar with how degraded vs intact RNA looks.

          It looks pretty good to me, nice sharp peaks and not much smearing. I think these samples would get a RIN of ~8 if the BA could assign a value. It is almost certainly just fine for library prep.
