  • Next-generation tutorial

    Hi everyone!

I am a new biochemistry graduate student. I have started working with next-generation sequencing. During my undergraduate studies, I learned the Sanger method very well but not the former. I have looked at google and youtube for starting tutorials (even Illumina), but I am still confused. For example, I am confused with adaptors.

Does anyone know of an online tutorial video?

Thank you very much for your time.

Someone posted a thread about a course on https://www.edx.org/ called PH525x Data Analysis for Genomics. You should still be able to sign up for it. If you do, then watching the "Next Generation Sequencing Technologies" lectures in week 2 will probably give you a good idea about what's going on. If I recall correctly, the videos give a bit more info than your typical intro that's on YouTube.

As far as info about adaptors, it doesn't go into an extremely rigorous explanation, but I think it's good enough to get the idea from what I remember.

      Not a video but a PDF: http://tucf-genomics.tufts.edu/docum...q_Adapters.pdf

This one is a little dated but virtually everything is still the same:

This course contains narrated presentations and video-based lab modules from the Genome Analyzer Boot Camp developed jointly by The Broad Institute and Illumina and held in February 2010. In this online version of the Boot Camp sessions, you’ll get an in-depth look at the chemistry and workflows for the Genome Analyzer. You’ll learn tips and tricks as well as best practices developed by the Broad to achieve even higher performance from the Illumina Genome Analyzer.

          Current Approaches to Protein Sequencing
          by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

          Strategies for Sequencing Challenging Samples
          by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
