  • What is the current state of sequencing?

    Hey all,

It's been several years since I last logged in and several years since I've been involved in sequencing. I'm curious how things have progressed in the last handful of years. Is there any good recent review article, blog, or equivalent that discusses how much progress has been made in recent years? Like, are there any new technologies, how much have prior technologies such as Illumina progressed regarding length of reads, number or reads per lane, cost for sequencing, etc? Any big improvements with respect to analyzing data sets?

One could easily argue I am being lazy in asking this question; I certainly don't expect anyone to spend a long time replying but if anyone has good links I would really appreciate it. Thanks!

Elaine Mardis' annual talk at the NHGRI's Genome Research Series is worth watching for a capsule intro to the latest in NGS tech. I am linking talk from 2016 since I can't find one from 2017.

Clive Brown's talk from London Calling conference gives a more focused update for Nanopore.

This thread has an excellent re-cap of all tech (see the post at end): http://seqanswers.com/forums/showthread.php?t=74469

Field guide to NGS sequencing (slightly out of date but still useful).

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
