1. Is there an upper size limit of fragment binding to the Illumina flow cells?
2. Would this be the same for Illumina GSII and for HS2000? 3. Can the Illumina protocol used for cluster formation be modified with respect to synthesis time? 4. If shorter fragments than the desired 1.5 kb get (unadvertedly) amplified too, would we obtain mixed clusters of short and long bridges that cannot be disentangled? 5. Did you ever try two or even more rounds of size selection by cutting from gels to avoid problems with shorter reads accumulating during cluster formation? |
