  • SOLiD 5500 fragment library preperation

    Hi all,
    I have a question regarding the fragment library prep for the SOLiD 5500 machine. In the initial shearing of the DNA with the covaris, ABI has added the inclusion of a shear buffer to the sample. This was not used in the SOLiD4 protocol. Since the rest of the protocol is the same between the two different machines, how important is this buffer to the sequencing on the 5500? Will not using this result in the sequencing to fail?

    I will be doing exomes with barcoding, so I am not sure if this will make a difference...

    I constructed several libraries for a nimblegen capture without using it and got good sequencing results.
