i am a new member of this amazing forum, and be honest it helps me a lot! i would like to ask if someone can give me main points about 454 Vs Sanger!

The 454 compared to Sanger generates many more but shorter reads at a much reduced cost-per-base -- at least 1/10th the cost. There is no cloning bias for the 454. There are problems with long homo-polymer runs.
