  • Baits length for exome capture? (Nimblegen and Aglient)

    Hi guys

    Does anyone use NimbleGen and Agilent exome capture?

    I wonder how long baits are for two methods.

    Agilent one is more than 100 bp, noted in brochuer but I can't find NimbleGen's one.
    I think it would be shorter than Agilent one because hybridization temperature of Nimblegen is lower than Agilent one.

    agilent has 120bp of target; nimble gen has 60-90bp

      The fixed length of the Agilent baits (120-mers) means the design algorithm is rather simpler, but has the potential disadvantage that base composition will affect the hybridization kinetics of individual baits/targets. Nimblegen tell me their baits are all designed to be isothermal, i.e. should all have the same hybridization kinetics at the temperature used (47 degrees C) and so the baits are of varying length to allow for different sequence composition.

        NimbleGen Exome Capture

        Hi ,

        What is the typical yield of nimble gen exome capture after capture/PCR (The final PCR).
        I got around 4 ug. Do you think the capture worked?
