  • multiple cartridges in a single MiSeq run?

    Hi All,

    I wonder if any one has attempted to use more than one reagent cartridge in a single MiSeq run, in order to increase the total cycle number over 300?

    Hi,

    It's possible, the easiest way is to prise the lids off the two cartridges and use the SBS reagents (3 larger pots) from one to top up the other. You can peel back the foil and fill them up almost to the top, then Parafilm the pots and replace the lid.

    You need at least 26ml of each for 2x201 and 32ml for 2x251, when you start the run it will warn you that the number of cycles specified in the sample sheet is too large but you can choose to ignore. Any more cycles than this is probably pointless as the 3' quality will be too low... or you could wait for the 2x251 upgrade

    Remember to top up the buffer too!
