  • HiSeq2500 Read Length

When introducing the HiSeq2500 Illumina promised 2x250bp by 2nd half of 2013. On the specification page they still list 2x150pb. Anyone has information or experience regarding longer runs in rapid run mode?

Unless you absolutely need the throughput ...

MiSeq supports 2 x 300 bp runs with v.3 chemistry now.

Indeed we would need the throughput... We have a couple of MiSeqs in operation.

Among other things we are waiting for are "ordered flowcells".

        (Ordered flowcells + 2 x 250 bp runs) = a winning combination

Anyone heard and theoretical numbers coming from ordered flow cells?
It would make sense for Illumina to begin roll out of new chemistries on MiSeq first due to the cost implications for any teething problems. So I expect to see them in a v4 chemisty (a part of me thought/hoped the read number jump in v3 would be down to ordered clustering).
Does anyone know if the new low diversity RTA will make it to HiSeq too?

James covered the patterned flow cell in a recent blog post. He is speculating about doubling the reads and >1Tb per run on HiSeq2500.

              Current Approaches to Protein Sequencing
              by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

              Strategies for Sequencing Challenging Samples
              by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
