  • Data normalization for whole transcriptome

    Dear all,

    I am new in this field, I would like to know what is data normalization in whole transcriptome. Please help me in this regard.

    Normalization is needed when you want to compare two libraries of RNAseq. Essentially you would want to know if the number of reads mapped to a gene in two libraries is different (or significantly different) but if you get more reads in one library more than in the other because you had more reads in the entire library the change in read counts is expected. The first step is to normalize the two libraries to have comparable values for each gene. You can read more about normalization methods in http://bib.oxfordjournals.org/content/14/6/671.long
