  • how to detect ori of plasmid

    hi, I want to predict ori of a plasmid. but it sems there is not a good method for plasmid. we can use GC skew to find ori of genome. is it available for plasmid.
    In addition, I found it is vary slowly to open up SEQanswers web in china. does anyone have the same question?

    Moving to ....um...general?

    You can guess at the origin of replication as the coverage may be higher there if you were to sequence a dividing population. I've seen this in bacterial datasets. I would guess that unless the plasmid is very large that you'd have a hard time detecting the overabundance of replication near the ori.

      thanks. my plasmid in a bacteria is only 11k. the relation between coverage and ori is interesting. could you explain it. why it happened?
