  • Ion Proton S5 Chips - 540

    Hello all, I am interested in purchasing the new Ion instrument, the S5 Prime, and was wondering if anyone has worked with the 540 chips before. We previously had an Ion Proton and used the P1 chips, and have been told the 540 is the upgrade to this chip, however I have not been able to find technical information. I know how many reads each chip claims it can produce, but since there are many variables to obtaining that max output, I am more interested in comparing how many beads I will have per chip, if the number of wells are the same, etc. Any help would be appreciated, thanks in advance.

    There are three run reports publicly(*) available for the 540 chip here:


    The "download" refers to just the PDF reports, but if you wish to download the datasets, you can use the clickable links provided in the PDF.

    For example, in the NA12878 run, an 89% loading resulted in 131450564 loaded wells. That would make the 540 in equal terms with the PI, at around 148 million wells.

    Overall it seems very similar to a nice, clean run done with a PI on the Ion Proton.
