  • TruSeq DNA Single Indexes Sets: How many samples on one lane

    Hi

    I'm new to Illumina sequencing and would like to run a few tests on a MiSeq platform using a TruSeq DNA Nano kit.

    As far as I understand, the Library Prep Kit will allow me to prepare 24 samples, and one of the DNA Single Indexes Sets (A or B) will give me 12 adapters, 24 samples and 48 preps. Does this mean that I can pool 24 samples on one MiSeq lane, and run two lanes? Or am I getting something wrong here? Is there a table with combinations to use for 24 samples on one lane? The library prep guide does not seem to provide this info.

    You can multiplex up to 24 samples/libraries provided that each has different index. For low pooling (less than 6) you need pay attention to index sequences to maximize base diversity (https://support.illumina.com/content...0041074-00.pdf)
