  • addSolexaReads.perl error

    Hey guys,

I'm trying to use the command addSolexaReads.perl to add the rest of my fastq reads to an assembly made with Newbler, but I keep getting the error "-ace must be specified." I think I'm including all of the correct files for the addSolexaReads.perl command but still keep getting that error.

Has anyone come across this before or might know where the problem is coming from?

    Thanks!

    Syntax has changed

The addSolexaReads.perl syntax changed around consed 23.

    Old syntax:
    Code:
    addSolexaReads.perl your_ace_file.ace your_fileoffilenames.fof your_fasta.fasta
New syntax, where you explicitly specify the options:
    Code:
    addSolexaReads.perl -ace your_ace_file.ace -fastqfof your_fileoffilenames.fof -fasta your_fasta.fasta

This video tutorial demonstrates how to align reads generated by Ion Torrent, 454, or Illumina sequencing to a reference sequence using consed. (Note that the…

      Current Approaches to Protein Sequencing
      by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

      Strategies for Sequencing Challenging Samples
      by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
