  • How should I compute the sampling depth or cells when I use pacbio RNA sequencing?

I want to do the RNA sequencing using pacbio SMRT, I have got the transcriptome information, but I don't know how many cells should I use ? And what the sampling depth is enough?

    Thank for any adviceO(∩_∩)O~

It depends on your goal and the organism's clade, of course. Are you doing novel transcript discovery from a well-known organism, for example? And what kingdom is it in?

Thanks for reply@Brian BushnelllO(∩_∩)O~ Yes. I want to do the novel transcript discovery and to accomplish one species's annotiation....

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
