  • emPCR on thermal cycler

    Hi Everyone,

I know that the Roche manual says that the emPCRs can sit at 10 degC on the thermal cycler for up to 16 hours. I was wondering if any one has any experience with leaving them on for longer?

I have started emPCRs and put them on the thermal cycler on Friday afternoon and left them until Monday morning when I started the breaking. I suppose this could make a difference, but I could not see any difference in my bead recovery rate or the run data when compared to other runs of the same samples.

I haven't left them for as long as a weekend, but I've left them 20 hours or so with no problems.

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
