  • What happens try to amp/lin/block/hyb paired end libraries on a single read flowcell?

Are the adapters the same or will this make a difference. Thanks

  • #2
Libraries that are prepared as paired-end libraries, will work on single-read flowcells without a problem.

What about the other way around? Could libraries prepared as single-read libraries (eg. with the small RNA sequencing adapters & primers) be run on paired-end flowcells?

It looks to me like the adapter sequences are different, and that it wouldn't work the other way around.

      Any thoughts?

No, libraries prepared with single read kits are not compatible with paired end cluster generation kits and flowcells.

All right, thanks so much! We'll have to go with single reads then.

            Strategies for Sequencing Challenging Samples
            by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
