Has anyone had issues with the kapa library quantification kit? I use it often and recently noticed I'm getting high ct values even in my control libraries. Do you leave the kit to thaw to room temperature for 30mins or do you leave it out longer? Recently I prepared a qPCR plate but the qpcr instrument was in use. I placed the plate in the fridge for 1 hr and placed on the qpcr when it freed up. The ct values were higher than expected in my controls but fell within my standard curve. Is this a phenomenon of the kit unhappy at 4degrees? Or is it something else? has anyone else had any issues with the Kapa library quantification kit they can share? I think it's a great kit but not without its issues that most people don't discuss and kapa tech support isn't awfully helpful in this regard either.

We recommend thawing our reagents completely while ensuring that they are protected from light. In addition, we generally recommend running the set-up qPCR plate as soon as possible.
