  • End repair of TruSeq RNA and TruSeq mRNA stranded

    Hi there,

    I have a question about the Library constranstion of TruSeq RNA and stranded mRNA prep kit.
    In the TruSeq RNA, reverse transcripted second strand cDNA is subject to End repair processing, but in the TruSeq stranded mRNA, that is not subject to End repair.
    Why the second strand cDNA that is synthesized by the TruSeq stranded mRNA kit is not required the end repair processing?

    Thyey have modified the enzyme mix for second strand synthesis so end repair also is done at that step. This is to simplify and speed up the process.

      Thanks for reply.
      You mean the enzyme mix contains T4 DNA pol and T4 PNK, isn't it?
