  • Unzipping TGZ file always halts

I'm using 7-zip to decompress/unzip my fastq files after downloading them to my machine, but it always halts at 68% (with an error that only names a single fastq file, but no further information about it or what went wrong).

Is this a common error or should I be concerned about the contents of that file?

    Any insight is greatly appreciated.

Is this happening with all files? If it is only one file then it is possible that the file that you are downloading is corrupt at source (or the corruption may be happening after you download).

By chance, are you using 7-zip on 32-bit machine?

Actually most of the NGS analysis tools will read compressed files. No need to uncompress.

      Thanks for the response.

I'm downloading the files from Galaxy, they are being stored by my institution. I don't think the files are corrupt, but it has occurred to me that the compressed file may be incomplete/ aborts the download due to large data size.

      I have a 64-bit machine.

Good to know that I can use compressed files during analysis, but the file I am trying to use has all of my fastq files, from all samples and treatments, so I would need to decompress it so I can access each library individually. Perhaps I should try downloading fewer samples at a time.

It is possible that 7-zip is writing the temp files to appdata/temp directory and filling up your C drive (or where ever your temp partition is). You should be able to see that happen.

Essential Discoveries and Tools in Epitranscriptomics
          by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist...

          Current Approaches to Protein Sequencing
          by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...
