  • Ideal RIN values for Soybean RNAseq !

    Hi

    Anyone know what should be the ideal RIN values for Soybean leaf, root and nodules.

    What should be Minimum RIN good for RNAseq ?

    Recommended RIN for RNA-Seq

    RNA Integrity Numbers should generally be >= 8 for RNA-Seq. I believe this threshold is recommended by the manufacturer of the BioAnalyzer (Agilent?). Apparently some labs use 7 as a threshold, but any paper I have read on the subject has used 8.

    Personally, I think close inspection of the electropherogram is more informative than the RIN. I have seen several low RIN's on electropherograms that had little to no background noise (i.e., indicative of low degradation) and all the important rRNA peaks at or close to the recommended ratios. rRNA ratios can vary with tissue/species. Moreover, a low 28S peak may not necessarily be indicative of universal RNA degradation, but can depend on physiological differences among tissues.
