  • how many chimera could be generated by circulation process of mate-pair preparation?

Im currently thinking to apply mate-pair library in my metagenomic project on cellulose-degrading microorganisms using Illumina Hiseq 2000 platform. but i was told the chimeric ratio could be as high as 20-30% in the circulation process of mate-pair library ! its a big concern for me but could not find much information in the papers?

So I would seek answers to these questions:
First, what is the chimera ratio for common circulation process of mate-pair preparation? is it matter with the insert size?

Second, what could be the chimera ration for metagenomic mate-pair preparation? in other words, how much more chimera could generate if DNA from different microorganisms is mixed together for the circulation process? Any experience on that?

Third, could anyone provide some reference in which the above questions were addressed with synthetic data? or if you have some experience on this issue could you share some data of yours?

Essential Discoveries and Tools in Epitranscriptomics
    by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist...

    Current Approaches to Protein Sequencing
    by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...
