  • Question (or request) about oligo half life at RTP?

    Hi,

    I can't seem to find info or references for the following question:

    What is the half life of PCR primers at RTP?


    I know you're supposed to store them at -20, and I know sometimes people use them after thawing them out for x hours, etc. etc. However, what I want to know is precisely how long you can expect PCR oligos to 'survive' at RTP, and what factors affect survival?

    Anyone know, or know someone who would know, or know a mailing or forum where someone who would know would be?

    Are there any papers on this?

    Anyone wan't to do a small study for me? ;-)

    PCR efficiency after using oligos thawed out 1h, 2h, 5h, and 10h ...

    In presense of little EDTA to ensure lack of heavy metal ions or Mg, at physiological conditions (pH 7-8) in absense of non-specific nucleases like phosphodiesterase well purified oligonucleotides should be quite stable. Products of oxidation or apurinization take long time to accummulate in siginificant amounts.
