we designed a custom Sureselect bait for ~0,8Mb (using which we want to sequence 200-300 patients samples on SOLID 5500xl).
Unfortunately Agilent does not have protocols for pre-capture barcoding and pooling. So instead they suggest to capture and then barcode, which is not efficient and too expensive. TargetSeq (Lifetechs version of Sureselect) is also too expensive and comes in huge quantities (96 minimum, ~ USD 26k).
Do you have an idea /hints for barcoding, pooling (e.g. 48 samples9 and then sequence capture using Sureselect for SOLID 5500xl?
