  • QIAGEN MIDI/Minispin Kit

    Hi All,

    I'm not sure if this is the best place to ask, but i'll ask anyway.

    So I was extracting DNA from Serum samples and proceeded using the QIAGEN Kit, after spinning the filters with AW1 and AW2 (both wash buffers), I accidentally added SDS to the column/filter instead of Ethanol. I found out about this right after i spinned the columns through with SDS and immediately added more Ethanol (what I was supposed to add) and spinned through it again. I was planning to use the elution to run PCR and DNA analysis, but I was wondering if the addition of SDS would make this DNA useless or interfere with it too much.

    ps. I added a small amount of SDS already along with the sample as part of starting material.

    Any help would be greatly appreciated.

    What % SDS? My guess is you would have lost a lot of your DNA. Did you save the flow through?
