  • Priming Flongle Flowcell

    Hi!
I hope you are very well, what protocols are there for priming the Flongle Flowcells?
    Thanks.

Well, there's the official ONT protocol which requires special tips and very careful standard plunge pipetting in order to not damage pores, and there's the negative-pressure gentle loading protocol which is more similar to the SpotON port loading method (search for "gentle loading") on the ONT community.

        Hello!
Regarding your question, Flongle flow cells are compatible with both R9.4 and R10.3 chemistry, and the protocol for priming them depends on the specific chemistry you are using.
For R9.4 chemistry, the Flongle flow cells can be primed using the protocol for priming MinION flow cells, which involves flushing the flow cell with running buffer and then washing with loading buffer. The detailed protocol can be found in the MinION flow cell user guide, available on the Oxford Nanopore Technologies website.
For R10.3 chemistry, the priming protocol is slightly different and involves priming the flow cell with EP buffer and then washing with loading buffer. The detailed protocol for priming R10.3 Flongle flow cells can also be found on the Oxford Nanopore Technologies website.
It is important to carefully follow the priming protocols for your specific chemistry to ensure optimal performance and to prevent damage to the flow cell.backrooms game

As far as I'm aware, Flongle flow cells have never been available for R9.4 nor R10.3. R9.4 was an earlier pore technology that stopped being used by ONT around 2019. R10.3 I'm less sure about. It might have been an earlier R10 technology, but was never offered on Flongle (except possibly to a sprinkling of labs for early-access testing).

Flongle flow cells are currently available on the ONT store in R9.4.1 and R10.4.1 pore formats ($810 USD for 12 flow cells).

            Current Approaches to Protein Sequencing
            by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

            Strategies for Sequencing Challenging Samples
            by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
